Research Interests:

Our laboratory is interested in understanding the epigenetic regulation of chromatin structure and gene expression programs during development, differentiation and disease initiation and progression. Compartmentalization of chromatin into active and silent domains is important for maintaining normal gene expression profiles. Our studies have shown that the insulator binding protein CTCF delineates the active and silent chromatin domains and that the loss of CTCF binding results in spreading of repressive chromatin, leading to gene silencing. Currently, we are investigating the role of transcription factors and epigenetic features that are involved in maintaining normal chromatin architecture and the anomalies that lead to changes in transcriptional program.

Another focus of our laboratory is to elucidate the mechanisms underlying gene expression alterations induced by environmental pollutants. The mutagenic potential of various environmental pollutants including several heavy metals are very low and do not correlate with their carcinogenic potential. Our studies show that nickel, an environmental carcinogen, could alter gene expression by disrupting chromatin domain structure. Currently, we are investigating the alterations to the epigenomic landscape induced by environmental carcinogens and their functional consequences.
