High Throughput Biology Laboratory Instruments & Facilities

NYU Langone’s High Throughout Biology Laboratory provides researchers with access to liquid handling automation and high throughput screening instruments.

Tissue Culture Room

Our tissue culture room features the following equipment:

  • 3 BSL 2 Biosafety cabinets
  • 4 37˚C Panasonic CO2 incubators for mammalian cell culture
  • a 25˚C incubator for Drosophila cell culture
  • an automated cell counter, plate centrifuge, inverted microscope, and 37˚C water bath

High Content Screening and Cell Imaging

Our laboratory uses advanced technologies to conduct high content screening and cell imaging.

Thermo Scientific ArrayScan High Throughput Imaging

Our Thermo Scientific ArrayScan VTI is a fully automated high content and high throughput cellular imaging microscope capable of imaging glass slides and high density microplates. This instrument has a 45-plate capacity hotel and a Thermo Scientific Catalyst Express robotic arm that feeds plates into the instrument for walkaway operation.

This microscope features the following objective lenses, listed according to magnification, numerical aperture (NA), long distance (LD) working capacity, and optical correction:

  • 2.5× (0.075 NA) Plan-NeoFluar
  • 5× (0.25 NA) Fluar
  • 10× (0.3 NA) Plan-NeoFluar
  • 10× (0.45 NA) Plan-Apochromat
  • 20× (0.4 NA) LD Plan-NeoFluar
  • 40× (0.5 NA) LD A-Plan

Fluorescence capabilities of the Thermo Scientific ArrayScan VTI include an automated excitation wheel containing optimal filters for the following:

  • broad blues [365(50)/535(45)]—Hoechst dyes, 4′,6-diamidino-2-phenylindole (DAPI) dihydrochloride, coumarins
  • greens [475(40)/535(45)]—fluorescein isothiocyanate (FITC), green fluorescent protein (GFP), fluorescein, Alexa Fluor® 488
  • orange reds [535(35)/590(35)]—tetramethylrhodamine (TRITC), rhodamine, cyanine dye (Cy®3), Alexa Fluor® 546
  • deep reds [575(25)/640(30)]—Texas Red®, Alexa Fluor® 594
  • long reds [630(50)/695(55)]—cyanine dye (Cy®5), DRAQ5, Alexa Fluor® 647

The system also features automated emission and dichroic rotation containing three dual band-pass filters and one triple band-pass filter.

Thermo Fisher Scientific EVOS FL Auto Cell Imaging System

Our laboratory also features the Thermo Fisher Scientific Invitrogen EVOS FL Auto Cell Imaging System, a benchtop bright-field, phase-contrast (PH), and fluorescence microscope with an automated stage for rapid imaging and analysis of cells and tissue sections. The instrument is capable of imaging from glass slides and 6- to 384-well microwell plates.

It features image overlay and composite analysis, as well as cell- and colony-counting capabilities.

This microscope features the following objectives, listed according to magnification, NA, working distance (WD), long working distance (LWD), PH, and optical correction:

  • 2× (0.06 NA, 5.1 mm WD) Plan-Achromat
  • 4× (0.13 NA, 16.9 mm WD) Infinity Plan-Fluorite
  • 10× (0.25 NA, 9.2 mm WD) LWD PH Plan-Achromat
  • 20× (0.4 NA, 3.1 mm WD) LWD PH Plan-Achromat
  • 40× (0.65 NA, 1.6 WD) LWD PH Plan-Fluorite

This instrument also features the following LED light cubes:

Plate Readers

NYU Langone’s High Throughout Biology Laboratory provides researchers with various plate reader technologies.

Filter-Based EnVision Multilabel Plate Reader

We offer the filter-based PerkinElmer EnVision® Multilabel Plate Reader, with the following features:

  • absorbance and colorimetric analysis
  • fluorescence intensity analysis
  • AlphaScreen® and AlphaLISA®
  • luminescence (luciferase, chemiluminescence)

EnSpire Multimode Plate Reader

We offer researchers the opportunity to use the PerkinElmer EnSpire® Multimode Plate Reader, a quadruple monochromator-based multimode plate reader with the following features:

  • absorbance and colorimetric analysis
  • fluorescence intensity analysis

LI-COR Odyssey Sa Infrared Imaging

We also offer the LI-COR Odyssey® Sa, an infrared imaging system that features the following assays:

  • far-red fluorescent secondary antibodies or DNA dyes
  • In-Cell Western
  • protein and DNA labeling

Liquid Handling and Automation

Our laboratory features several liquid-handling systems.

EL406 Microplate Washer Dispenser

The BioTek EL406 Microplate Washer Dispenser is a compact system that can dispense or aspirate into microplates (96- or 384-well density). This system includes the following functions and protocols:

  • cell fixing and staining
  • enzyme-linked immunosorbent assay
  • media or reagent exchange
  • liquid dispensing

Washing, including dispensing and aspirating, is conducted with a 96-tube manifold with 96 stainless tubes. The volume range is 1 to 1,000 μL per well.

The BioTek EL406 Microplate Washer Dispenser offers a choice of a peristaltic or syringe pump.

A peristaltic pump is used primarily for expensive or limited reagents. The following volume ranges and minimum prime volumes are recommended:

A syringe pump is used primarily for dispensing cell media or buffers (inexpensive reagents) and has the following features:

  • 3 to 3,000 μL per well
  • selectable, in 1 μL increments
  • minimum priming volume of 12 mL

WellMate Dispenser

We also offer several Thermo Scientific Matrix® WellMate® Microplate Dispenser bulk liquid handling systems. Some are integrated with stackers for processing of multiple plates. Primary uses include cell and reagent dispensing in 96- and 384-well formats. Other plate formats are also available.

Standard bore tubing includes the following specifications:

  • 20 to 2,000 μL dispense volume
  • selectable in 1 μL increments up to 100 μL
  • selectable in 5 μL increments from 100 to 2,000 μL
  • minimum prime volume of 5 to 8 mL

Small bore tubing includes the following:

  • 5 to 200 μL dispense volume
  • selectable in 1 μL increments up to 100 μL
  • selectable in 5 μL increments from 100 to 200 μL
  • minimum prime volume of 5 to 8 mL

Formulatrix Mantis Microfluidic Liquid Dispenser

The Formulatrix Mantis is a low dead volume microfluidic liquid dispenser that can be customized for specific well dispensing of 0.1μL and up. Primary uses include optimization of transfection reagent, antibody titration, and statistical design of experiments.

Agilent Bravo Liquid-Handling System (384-pipet head)

Our laboratory has an integrated liquid-handling system consisting of Agilent Bravo (96- or 384-pipette head) Liquid Handler, Agilent BenchCel 4R Microplate Handler, Agilent PlateLoc Microplate Sealer, and Agilent Microplate Labeler enclosed within a class II biosafety cabinet to ensure sterile conditions.

Applications include sterile dispensing of cells, reagents, antibodies, and media exchanges, as well as plate labeling and library duplication. Cell and reagent dispensing is in 96- and 384-well formats, with volume ranges of 0.5 to 30 μL and 0.75 to 70 μL, respectively. Custom protocol development is available.

PerkinElmer JANUS MDT Varispan Automated Workstation

The PerkinElmer JANUS® Varispan Automated Workstation is a robotic liquid-handling instrument that uses a 96 Modular Dispense Technology (MDT) pipette head and 8-tip Varispan to transfer liquid to and from high-density microplates (96 and 384 wells). Custom protocol development is available.

Primary uses of the system include cherry-picking small interfering RNA hits, library reformatting, and reagent dispensing in high-density microplate formats with volumes of 0.5 to 235 μL.

Reagents and Supplies

Our laboratory is stocked with the following microplates, liquid handling supplies, assay reagents, and other items.

Available 96-well plates include the following:

  • black, clear bottom, tissue culture–treated microplates; Corning 3904
  • white, solid bottom, tissue culture–treated microplates; Corning 3917
  • white, clear bottom, tissue culture–treated microplates; Corning 3903

Available 384-well plates include the following:
