Interventional randomized double-blind parallel-group placebo-controlled multi-centre study to assess the efficacy safety and tolerability of Lu AF82422 in patients with Multiple System Atrophy
This is a phase II, interventional, randomized, double-blind, parallel-group, placebocontrolled, multi-centre study of the efficacy (slowing disease progression), safety, and tolerability of Lu AF82422 in patients with MSA.MSA is a progressive, fatal disorder characterized by autonomic failure and parkinsonismand/or cerebellar involvement. Neuropathologically, MSA is characterized by oligodendroglial inclusions of abnormally aggregated a-synuclein. No disease-modifying therapies are currently available; symptomatic treatment options are limited, and the therapeutic benefit of these therapies is often only transient. Thus, there is a critical unmet need for a treatment that can delay the progression in MSA. a-synuclein is considered a valid target for immunotherapies that aim to neutralize and/or clear toxic a-synuclein species, Lu AF82422 is a human IgG1 mAb that recognizes all major species of a-synuclein and is being developed as a treatment for delaying of disease progression in idiopathic PD and/or MSA.Nonclinical and clinical data of Lu AF82422 supports its potential as a safe and well tolerated treatment to reduce or inhibit seeding of pathological form(s) of a-synuclein, and potentially delay disease progression in MSA.

Intra-Individual Variability in Cognitive Performance as a Marker of Prodromal Disability in MS
The goal of this project is to evaluate intra-individual variability (IIV) in cognitive performance, measured across response time (RT) on a computer task, as a marker of potential cognitive decline. We currently identify a patient’s advancing neurological decline when it reaches the threshold for clinical detection of change. There remains an urgent need to identify patients before the onset of decline and disability.This project will be a prospective observational clinical study where we will recruit adults with early MS (n=60) into two groups. One group (n=35) will complete a cognitive assessment, a single 60-minute brain MRI combined simultaneously with 20 minutes of 2.0mA tDCS, and an optional gait assessment using a wearable motion sensor. A second group (n=25) will complete the cognitive assessment and optional gait assessments only.
Intradialytic Myocardial Study Stunning in Maintenance Hemodialysis Patients: A Multi-Center Observational Cohort Study
The purpose of this observational study is to prospectively investigate the frequency and risk factors for intermittent intradialyltic myocardial stunning (IDMS) in maintenance hemodialysis patients, to evaluate the prevalence of repetitive IDMS, and to investigate the role of cardiovascular autonomic control in IdMS pathophysiology using a battery of autonomic function tests during dialysis and on non-dialysis daysAll subjects will undergo pre- and intradialytic transthoracic echocardiography during a single hemodialysis session. There will be four substudies:• Substudy A—a subset of participants will undergo repeat scanning during a 2nd session to assess the frequency of repetitive IDS• Substudy B—a subset of participants will undergo intradialytic testing of autonomic function during a single hemodialysis session during the baseline echocardiography scanning• Substudy C—a subset of participants will undergo autonomic function testing on a non-dialysis day• Substudy D—a subset of participants will undergo myocardial sympathetic PET scanning in order to assess myocardial uptake of neuro-sympathetic hormones
