Microscopy Laboratory Electron Microscopy Technologies & Instruments

NYU Langone’s Microscopy Laboratory uses powerful electron microscopes to obtain high-resolution images of your specimens. Electron microscopy sample preparation is generally a drop-off service. However, if you prefer to perform your own imaging and are a new user, we can provide training. If you are an experienced user, we can offer—with approval—the materials required for your experiment. Electron Microscopy Technologies

Our in-house electron microscopy technologies include the following:

  • embedding using either epoxy or acrylate-based embedding resins at ambient or low temperatures
  • thin sectioning using a Leica Ultracut UCT microtome or Leica EM UC6/EM FC6 cryoultramicrotome
  • cryoultramicrotomy using a Leica EM UC6/EM FC6 cryoultramicrotome
  • immunolabeling following pre- or postembedding protocols
  • Tokuyasu immunogold labeling of ultrathin cryosections
  • high-pressure freezing using a Bal-Tec HPM 010 (located at the New York Structural Biology Center), and freeze substitution by a Leica EM AFS2 automatic freeze-substitution system
  • negative staining of viruses, bacteria, and macromolecules
  • rotary shadowing using a BOC Edwards Auto 306 evaporator for DNA, RNA, and macromolecules
  • electron tomography data collection using either a Philips CM200 or an FEI Tecnai F20 transmission electron microscope (TEM) (located at the New York Structural Biology Center)
  • three-dimensional electron microscopy using a newly purchased Zeiss Gemini 300 field-emission scanning electron microscope (SEM) with Gatan 3View® or an FEI Helios 650 DualBeam SEM (located at the New York Structural Biology Center)
  • correlative light and electron microscopy

  • Philips CM12 TEM with Gatan 4K × 2.7K digital camera
  • Philips CM200 TEM with cryoholder, and Gatan 2K × 2K digital camera capable of tomographic data acquisition

  • Leica EM UC6/EM FC6 cryoultramicrotome
  • Leica EM FCS cryoultramicrotome
  • Leica glass knife maker
  • BOC Edwards Auto 306 evaporator
  • Gatan Solarus advanced plasma system
